Abstract 1319 Impact of ATG-5 Knockdown on aUPR and Cell Necrosis in Breast Cancer Cells Treated With BHPI
The small molecule 3,3-bis(4-hydroxyphenyl)-7-methyl-1,3,dihydro-2H-indol-2-one (BHPI), induces cell necrosis via estrogen receptor α (ERα) in breast and ovarian cancer cells. BHPI induces necrosis by hyperactivating the anticipatory Unfolded Protein Response (aUPR) which results in near total protein synthesis inhibition, cellular swelling, and eventually causes depletion of cellular ATP. BHPI also inhibits autophagy, as seen by decreasing production of both LC3-I, LC3-II, and Beclin-1 via western blot.
