Abstract 1145 Adenylyl Cyclase 2 Negatively Regulates the Blood-Brain Barrier after Intracranial Hemorrhage
Intracranial hemorrhage (ICH) is a rarer, but more deadly, form of stroke that occurs when a blood vessel bursts in the brain parenchyma. There are currently no treatments for ICH besides surgical intervention, stemming from a lack of viable therapeutic targets for the repair of the blood-brain barrier (BBB) following hemorrhagic injury. Adenylyl cyclase 2 (Adcy2) was identified as a gene upregulated in mice lacking endothelial-cell derived laminin gamma-1 that exhibit a phenotype of BBB leakiness; BBB instability increases the risk of hemorrhagic event, and we hypothesize that Adcy2 aggravates hemorrhagic stroke by weakening BBB integrity, therefore preventing efficient vessel repair and promoting excessive extravasation of immune cells at the site of injury.
