Abstract 2899 Optimization of Encapsulation and RNA Preservation of T Cells for Non-Microfluidic scRNA-Seq
High-throughput single-cell RNA sequencing (scRNA-seq) has revolutionized our understanding of RNA molecules and has enabled quantitative analysis of gene expression. In the context of T cells, scRNA-seq can be used to analyze the T cell receptor (TCR) repertoire, which provides insights to T-cell antigen specificity. Despite its potential, the widespread application of scRNA-seq is often constrained by its high cost and the susceptibility of RNA degradation in the sample during processing. To address these limitations, we focused on optimizing the encapsulation efficiency and RNA preservation in a non-microfluidic scRNA-seq method called particle-templated instant partition sequencing (PIP-seq), using T cells in the experimental system.
