Abstract 1251 Novel Methionine Aminopeptidase Inhibitor of Mycobacterium is Not Cytotoxic to Mammalian Cells

accumulation and hepatotoxicity caused by the high-fat diet.In-vitro experiments showed that Rhy effectively suppressed the ox-LDL uptake by macrophages and subsequent foam cell formation by enhancing the cholesterol efflux transporters ABCA1, ABCG1, and the nuclear transcription factor LXR-a.The cell transfection experiments using LXRa siRNA showed that the enhanced cholesterol efflux by Rhy is via LXR-a activation and the increased LXR-a activation was reverted when AMPK was inhibited by specific inhibitor, and by transfection with AMPKa1 siRNA.Conclusion: Rhy has a strong anti-atherosclerotic effect, possibly through the AMPKa1-LXRa-ABCA1/ABCG1 pathway by promoting cholesterol efflux from macrophages and inhibiting foam cell formation.
