Abstract 1917 A solution NMR based approach to characterizing ASCC2-CUE specificity for K63-linked polyubiquitin chains in DNA alkylation damage repair
DNA alkylation is a mutagenic and potentially cytotoxic DNA modification that entails the addition of an alkyl group to DNA bases via nucleophilic substitution. DNA alkylation can occur randomly through endogenous sources, resulting in up to 20,000 lesions per day in a single cell. One way in which these lesions are repaired is via the ALKBH3-ASCC repair complex, which is localized to regions of alkylation damage in a K63-linked polyubiquitin-dependent manner. In the complex, ALKBH3 acts as a dealkylating enzyme, while the ASCC3 subunit has helicase activity.
