Abstract 3035 Cellular conditions shape CRISPR-Cas specificity and mechanism
Cas9 and Cas12a are immune effectors of bacterial CRISPR-Cas defense systems and have been adapted for genome editing and other biotechnology tools. Cas effectors have been the subject of extensive investigation both in vitro and in genome editing experiments, providing detailed insight into the mechanism and specificity of these important enzymes. However, many of these studies have been performed in conditions that are not native to bacteria, especially as relates to divalent metal ion concentrations.
