Abstract 2902 The Role of Klotho-alpha in the Stabilization of FGFR and Its Effects on Longevity through FGF23 Pathways
Depending on its location and the cell to which it binds, Klotho can cause a vast series of effects related to the slowing of aging. Klotho-alpha acts as a co-stabilizer for FGF23 receptor through its binding between FGF23 and both tyrosine kinases which form FGFR. Within the kidneys, the FGFR-Klotho-alpha complex and FGF23 are key regulators of phosphate and Vitamin D production through the suppression of 1α-hydroxylase expression and the promotion of 24-hydroxylase transcription. Within the bone matrix, Klotho's effect on aging is seen through its interaction with FGF23 again, which plays a critical role in mineral ion homeostasis, thus regulating bone mineralization and protecting against decay.
