Abstract 2117 Potential of established and novel antibacterials against Acinetobacter baumannii gyrase and topoisomerase IV
Acinetobacter baumannii is a health-care associated infection that has major therapeutic challenges due to extensive antibacterial resistance. Most A. baumannii infections are resistant to fluoroquinolones (FQs), a class of gyrase/topoisomerase IV poisons. Therefore, we assessed the effects of two novel antibacterials, gepotidacin (a triazaacenaphthelene) and zoliflodacin (a spiropyrimidinetrione), on the activities of A. baumannii gyrase and topoisomerase IV. FQ-resistance is associated with single-amino acid mutations in the proteins that disrupt the interaction between FQs and the enzyme.
