Abstract 2622 Characterizing the stabilization effects of Aurora Kinase A on ubiquitin mediated degradation of c-Myc
The Myc gene family comprises three highly conserved transcription factors, c-, N-, and L-Myc, known to be oncogenic drivers in many cancers. Dysregulated expression and increased stability of Myc are estimated to drive one-third of all cancers making Myc an attractive anticancer target. Because Myc lacks druggable pockets it may be necessary to disrupt its protein-protein interactions to modulate Myc function or stability. Myc stability is regulated by growth factor signaling, where phosphorylation of residue Threonine 58 within the Myc phosphodegron sequence triggers Myc ubiquitination by the SCFFbxw7 ubiquitin ligase and subsequent degradation.
