Abstract 2429 Effects of phosphatidylserine on the kinase activity of epidermal growth factor receptor
Activity of membrane proteins, such as the epidermal growth factor receptor (EGFR), is affected by several lipids in the plasma membrane such as the phosphatidylserine (PS). Reported studies of the role of PS on EGFR activity are more on a computational and molecular dynamic perspective. Here we investigate the interactions of PS with a poly-His-tag EGFR intracellular domain (ICD) in vitro. The His-tagged EGFR-ICD was incubated in liposomes decorated with a nickel-nitrilotriacetate head group-containing lipid (1, 2-Dioleoyl-sn-Glycero-3-{[N(5-Amino-1-Carboxypentyl)iminodiAcetic Acid]Succinyl} Nickel salt, DOGS-NTA-Ni) over a range of PS concentrations.
