Abstract 2017 Engineering a Minimal Synthetic Cell Cycle in a PURE Cell-Free Protein Synthesis System
Temporal regulation is a fundamental feature of all living systems. Mimicking and manipulating these natural dynamics is a key goal in synthetic biology. Therefore, tools for genetic temporal regulation are of essence, as they could be applied for synchronizing bioproduct manufacturing with growth phases, engineering biosafety killswitches, and streamlining cell-free design-build-test platforms. In this work, we aim to engineer a minimal module for synthetic cell cycle control by integrating transcriptional regulation with targeted protein degradation in PURE system.
