Abstract 1076 Creation of a new genomically recoded organism with a single non-degenerate stop codon
Genome engineering approaches have paralleled natural codon reassignment by using whole-genome replacement of synonymous codons to construct genomically recoded organisms (GROs). Previously, we replaced 321 TAG stop codons in the E.coli genome with synonymous TAA permitting release factor 1 deletion and the first GRO (C321.ΔA) with clean TAG “reassignment” for non-standard amino acids (NSAA). Additional codon reassignment would allow for genetic code expansion to combinations of NSAAs and test the limits of codon compression.
