Abstract 1639 A polyphosphate kinase for the chemo-enzymatic production of base-modified nucleoside 5′-triphosphates

Nucleic acid modifications play important roles in processes including regulation of gene expression and protection from degradation in both endogenous and therapeutic RNAs. Production of modified RNAs in vitro requires access to a suite of modified nucleoside 5'-triphosphates (NTPs). Most current strategies for NTP synthesis suffer from low yields or require costly phosphorylation reagents. We recently presented a chemo-enzymatic approach for producing base-modified ATP analogues. Adenosine or analogues thereof were first chemically phosphorylated to form adenosine 5'-monophosphates.

Abstract

Nucleic acid modifications play important roles in processes including regulation of gene expression and protection from degradation in both endogenous and therapeutic RNAs. Production of modified RNAs in vitro requires access to a suite of modified nucleoside 5'-triphosphates (NTPs). Most current strategies for NTP synthesis suffer from low yields or require costly phosphorylation reagents. We recently presented a chemo-enzymatic approach for producing base-modified ATP analogues. Adenosine or analogues thereof were first chemically phosphorylated to form adenosine 5'-monophosphates.
