Abstract 2416 Use of Reverse-Phase HPLC and SYBR Gold-Stained Mini Gels for Analysis of DNA Glycosylase Activity
DNA glycosylase activity is typically monitored by using radiolabeled or fluorescently labeled DNA followed by base-induced strand cleavage and denaturing gel electrophoresis. Fluorescently-labeled oligonucleotides are expensive, and radiolabeling is not possible at many institutions today. In addition, the equipment used to run large-scale polyacrylamide gels for nucleic acids is becoming difficult to find. The goal of this project was to develop a quantitative method for assaying the activity of uracil DNA glycosylases that could be carried out without labeling the DNA.
