Abstract 1938 Unveiling Human Inorganic Pyrophosphatase 1: Crystallographic Insights and Activity Studies for Anti-Cancer Target Development
Inorganic pyrophosphatases are essential, highly specific, ubiquitous enzymes that catalyze the hydrolysis of one molecule of pyrophosphate (PPi) to two molecules of metabolizable inorganic phosphates (Pi) in the presence of divalent metal cations. Inorganic pyrophosphatases are central to cell metabolism due to their key role in maintaining cellular phosphorus homeostasis by removing the PPi released during biomolecule synthesis, thus making biosynthetic reactions thermodynamically favorable. Upregulation and accumulation of human inorganic pyrophosphatase 1 (hsPPA1) has been associated with cancer cell proliferation, invasion, and migration, making it a promising enzyme target for potential prognostic biomarker and development of small-molecule anti-cancer agents.
