Abstract 1953 PI3K signaling in health and disease: new twists and turns
The phosphoinositide 3-kinase (PI3K)/AKT signaling pathway has been the focus of numerous studies over the past 30 years. We now know that hyperactivation of this pathway is common in cancer and the cause of a group of rare yet non-cancerous overgrowth disorders. However, a fundamental knowledge gap remains and hampers the effective use of targeted PI3K inhibitors in the clinic: we still cannot predict, let alone control, how biochemical specificity is encoded and subsequently decoded through activation of the PI3K/AKT pathway, in health or disease.
