Abstract 1458 Tracking the single-molecule inheritance of chromatin structure in purely reconstituted chromatin replication reactions
As DNA replicates, chromatin organization must be faithfully inherited by both daughter strands to safeguard cell identity. We have applied a strand-specific version of RASAM (replication-aware single-molecule accessibility mapping) to a fully reconstituted S. cerevisiae replication system. RASAM is a recently-published chemical method that utilizes bromodeoxyuridine (BrdU) incorporation to mark replicated fibers, followed by footprinting protein-DNA interactions with a non-specific adenine methyltransferase, then sequenced using PacBio long-read sequencing.
