Abstract 2528 An intrinsically disordered region of the FACT subunit, Spt16, regulates chromatin dynamics in controlling the assembly of transcription factors at the promoter for transcription initiation
Previous studies revealed that an intrinsically disordered region (IDR) of the Spt16 subunit of an evolutionarily conserved histone chaperone, FACT (Facilitates chromatin transcription), interacts with histone H2A-H2B dimer, and the Spt16 IDR mutations are associated with a number of cancers. However, the functional relevance of Spt16 IDR's interaction with histone H2A-H2B dimer in regulating chromatin and transcription has not been elucidated in living cells. Here, using genetic, biochemical and molecular biological methodologies, we have analyzed the role of SPT16's IDR in regulation of chromatin and transcription.
