Abstract 2283 Modulation of Super-Enhancer Activity in T-ALL JURKAT Cells Utilizing Synthetic Zinc Finger Proteins
Tight regulation of gene expression is mandatory for efficient cellular differentiation and the prevention of oncogenesis. When genes become dysregulated it can lead to differentiation blockades and abnormal cellular proliferation. A common avenue of gene dysregulation is the modulation of enhancer activity that leads to high-level, aberrant expression of oncogenes. A subset of T-cell Acute Lymphoblastic Leukemia (T-ALL) is characterized by an insertional mutation upstream of the T-cell Acute Lymphocytic Leukemia Protein 1 (TAL1) gene locus that creates a de novo binding site for c-MYB, a transcription factor, that in-turn recruits a multitude of other transcription factors to form a super-enhancer complex.
