Abstract 1518 A Binding Interaction Analysis of UHRF2 Mutants
Epigenetic mechanisms play a significant role in regulating gene expression. One of these mechanisms involves histone reader proteins recognizing histone modifications. UHRF1 and UHRF2 are histone reader proteins that are associated with the development of cancer. They are able to bind modified histones at specific DNA sequences, in turn down-regulating gene expression. Our research goal is to determine the specific interactions between UHRF2 and Histone 3 (H3) that are critical. To do this, we created a series of mutations in the TTD-PHD, TTD and PHD of UHRF2 and measured their ability to bind H3.
