Abstract 2938 Dissecting the Epigenetic and Transcriptional Dynamics of Somatic-to-iPS Reprogramming at Single-Cell Resolution
The process of somatic-to-iPS reprogramming leads to the invariable silencing of diverse somatic identities and the establishment of a singular pluripotent cell state (iPS). The molecular mechanisms by which diverse somatic programs become destabilized for a single pluripotency program to emerge remain poorly defined. By multiomic (scRNA&ATAC-seq) profiling a quarter of a million reprogramming cells spanning diverse somatic origins and sexes, we constructed a comprehensive epigenetic and transcriptomic roadmap of cellular reprogramming.
