Abstract 2573 Exploring the effect of O-GlcNAc on Pomc transcript stability
One in ten people will develop a pituitary adenoma in their lifetime. In the pituitary gland, as in the entire human body, proteins are constantly modified by the post-translational modification O-linked N-Acetylglucosamine (O-GlcNAc). Protein O-GlcNAcylation is associated with multiple cellular functions, from regulating transcription/translation to cell development and metabolism, and increases during periods of elevated glucose concentration, thereby acting as a nutrient sensor. In this study, we explore how O-GlcNAcylation contributes to increased pro-opiomelanocortin (POMC) expression in adenomatous corticotropic cells and subsequent elevated levels of adrenocorticotropic hormone (ACTH), a product of POMC cleavage.
