Abstract 2122 Evaluating interactions between the Rho GTPase TCL and Coronin2b using split-luciferase assays
TCL/RhoJ is an endothelial-specific member of the Cdc42 subfamily of Rho GTPases and plays a pathological role in tumor-associated angiogenesis. TCL possesses a distinctive N-terminus compared to other Cdc42 family members, which is essential for its localization to the plasma membrane. This localization is nucleotide-dependent, with GTP-loaded TCL residing at the plasma membrane and GDP-loaded TCL associating with vesicles. Using a split luciferase protein complementation assay, we identified an interaction between Coronin 2B and TCL, which may facilitate the tethering of TCL to vesicle membranes.
