Abstract 2849 The role of TMEM106B and PMP22 in various neurodegenerative diseases
TMEM106B is a type II transmembrane protein comprised of three key domains: N-terminal cytosolic domain, transmembrane domain, and C-terminal luminal domain. TMEM106B plays a crucial role in lysosomal function, maintaining lysosomal pH and trafficking, by interacting with microtubule-associated protein 6 (MAP6) and vacuolar-ATPase (vATPase). Dysfunction of TMEM106B can lead to various neurodegenerative conditions, including fronto-temporal dementia (FTD) and Amyotrophic Lateral Sclerosis (ALS). Importantly, TMEM106B's proteolytic processing of the C-terminal results in the formation of amyloid fibrils, which are implicated in these diseases.
