Abstract 1225 Expanding the toolkit: Synthesis and evaluation of 3,4-dihydroxyhydrocinnamic acid derivatives as mechanistic probes of extradiol dioxygenase activity
Extradiol dioxygenase (EDX) enzymes are essential protein catalysts for the breakdown of catecholic rings. This powerful chemistry is used in nature to degrade lignin-derived aromatic carbon and even make natural products, but a paucity of available chemical tools has limited our understanding of the breadth and depth of substrate space for these powerful enzymes. In order to investigate L-DOPA 2,3-dioxygenase from the biosynthetic pathway to the antibiotic, lincomycin and its homologous enzymes, we created a first-of-its-kind toolkit of 6-substituted catecholic derivatives.
