Abstract 2397 Polyphenon E significantly effects gene expression in DU145 cells
significant reduction of FZD6.Additionally, we found that either knockdown of SP1 or FZD6 decreases cell motility, as measured by Transwell assays. To confirm that reduced FZD6 expression results in less motile PDAC cells in vivo, we crossed a tissue-specific Fzd6 knockout mouse model to the wellknown, tumorigenic KPC model. We found that the loss of Fzd6 in PDAC cells dramatically inhibits tumor progression and metastasis rates.Mice lacking Fzd6 in their tumors had an increase in average survival time by 25% and reduction of metastases by 20%.Taken together, we believe we have identified a WNT4/WNT5A/FZD6 driven PCP pathway that makes PDAC tumors more motile, and results in increased PDAC metastasis and worse patient prognosis.
