Abstract 2862 Toward photocontrol of Src kinase

Bacteriophage T4 early gene products, including motB.1, are non-essential under laboratory growth conditions that remain still with unknown functions and may be involved in phagehost interactions.Translation of small heat shock genes in aand g-proteobacteria is controlled by the ROSE (Repression Of heat Shock gene Expression) element, a thermo-responsive RNA structure in 5' UTR which regulates the ibpA gene encoding for an inclusion body-associated protein. We demonstrate here that T4 early protein MotB.1 post-transcriptionally regulates the IbpA protein in Escherichia coli.Using an ibpA fluorescence reporter system that contains the mutation strains fused to the mCherry gene, we find that the overexpression of MotB.1 resulted in a 1.6-fold increase in the mCherry levels, indicating that MotB.1 affects the ibpA RNA thermometer.Furthermore, the mCherry level exhibited a significant increase in the sequence of mutation 1, 2, and 3, suggesting that gene expression rises as the Shine Dalgarno (SD) region becomes more accessible or a hairpin structure is less likely to form after the SD region.Mutation 4 presented an increase of around 1.3-to 1.5-fold compared to ibpA 5' UTR, and the deletion of cgt after the start codon did not result in a notable difference in the mCherry level. Thus, the marked difference in the mCherry level of mutation 3 is a consequence of the complete disruption of the ibpA 5' UTR, rather than the cgt deletion.
