Abstract 1808 The development of chemical probes of histone “reader” subunits of chromatin remodeling complexes
Histone posttranslational modifications are deposited by “writers”, removed by “erasers”, and recognized by effector proteins called “readers”. Reader proteins contain discrete domains that bind histone posttranslational marks, and are typically subunits of larger functional protein complexes. These chromatin targeting complexes are involved in genome maintenance and transcriptional regulation, in part by moving nucleosomes to regulate DNA accessibility. The mammalian SWI/SNF, or BAF, chromatin remodeling complex has been implicated as a therapeutic target in cancer, although the most effective way to target this heterogeneous multi-subunit complex is unclear.
