Flavonoid content of Pandan Anggor (Sararanga sinuosa Hemsley) andantioxidant activity during storage time

Pandan Anggor is a native species of the Papuan archipelago and remains extensively understudied scientifically. As a member of the Pandanus family, the fruit of Pandan Anggor shares characteristics with berries such as raspberries, blueberries, and blackberries. Berries are rich in bioactive components, including flavonoids, which possess various beneficial properties such as antioxidant, anti-inflammatory, and antitumor agents. Therefore, this study aimed to investigate the flavonoid and antioxidant activity of Pandan Anggor fruit during storage. The measurement of total flavonoid levels was carried out through a colorimetric method using an Ultraviolet-Visible (UV) spectrophotometer. Antioxidant activity test was determined by the IC50 values of 2,2- diphenyl-1-picrylhydrazyl (DPPH) and Ferric Reducing Antioxidant Power (FRAP) methods, while flavonoid types were identified through Liquid Chromatography – Mass Spectrometry/Mass Spectrometry Quadrupole Time of Flight (LC-MS/MS QTOF). The results showed that the highest total flavonoid levels were found on day five of storage. Specifically, total flavonoid content in fresh fruit water extract, fresh fruit ethanol extract, and fruit powder ethanol extract was 6.10%, 11.60%, and 14.30% dry basis, respectively. According to DPPH and FRAP, the antioxidant activity of the ethanol extract of Pandan Anggor fruit simplicia was the highest compared to others, with IC50 values of 7.92 ppm (DPPH) and 5.97 ppm (FRAP), respectively. This study showed that storing Pandan Anggor fruits for 5 days after harvest provided the highest levels of flavonoid and antioxidant activity. LC-MS/MS QTOF analysis showed the presence of iridin, luteolin-7- O-D-glucoside, undulatoside A, isoharmnetin-3-O-neohesperidoside, liquiritin apioside, quercetine-3-rhamnogentiobioside, viscumneoside VI and II.
