Anticancer Activity of Bis (4-carboxyanilinium) Selenate in Mouse Fibrosarcoma Cells

A BSTRACT Background: Bis (4-carboxyanilinium) selenate (B4-CASeO 4 ) is known to have antioxidant, antibacterial and antifungal activities. However, its anticancer activity is not yet studied. Materials and Methods: This study evaluates the anticancer activity and underlying mechanism of B4-CASeO 4 in mouse fibrosarcoma (WEHI-164) cells. Results: B4-CASeO 4 caused a concentration-dependent decrease in the proliferation of WEHI-164 cells, which was correlated with a decrease in clonogenic survival. Cells treated with B4-CASeO 4 resulted in enhanced intracellular reactive oxygen species generation and a decrease in mitochondrial membrane potential. Moreover, cell death measured by sub-G1 analysis by flow cytometry showed an almost two-fold increase. Treated cells also resulted in increased level of cleaved Caspase-3 suggesting apoptotic mode of cell death. Conclusions: Collectively, these results suggest the involvement of oxidative stress mediated apoptotic death by B4-CASeO 4 in WEHI-164 cells.
