Determination of seven components in Huatuodou by HPLC based on quantitative analysis of multi-components by single marker (QAMS)

Abstract Huatuodou is the dried mature seed of Calonyction muricatum (L.) G. Don, a plant of Convolvulaceae, which was first recorded in ‘Jiuhuang Bencao’. Studies have shown that Huatuodou have obvious anti-inflammatory effects, and concluded that neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, isochlorogenic acid A, isochlorogenic acid C may be the pharmacodynamic substances. Its quality has not been extensively evaluated in spite of its anti-inflammatory. The main objective of this study was to establish a quality control method of one-measurement-multiple-assessment method (QAMS) for Huatuodou, and to investigate the accuracy and applicability of this method by comparing it with the external standard method. In this study, the contents of seven components, namely chlorogenic acid, neochlorogenic acid, cryptochlorogenic acid, caffeic acid, isochlorogenic acid A, isochlorogenic acid B and isochlorogenic acid C, were simultaneously determined by high performance liquid chromatography (HPLC). The relative correction factors (f S/i ) of the other six components were calculated using the multipoint correction method with chlorogenic acid as the internal reference, and the differences between the results of the QAMS method and those of the external standard method were compared to verify the accuracy of the QAMS method. The results showed that all the f S/i values had good applicability (RSD<5%) under different column temperatures, column, flow rate and different instruments, and there was no significant difference between the quantitative results of the seven components in Huatuodou from 10 origins under different quantitative methods. The applicability and feasibility of the QAMS method established in this study were evaluated to be favorable for the quality control of Huatuodou.
