Evaluation of the parameters of subchronic toxicity parameters of an antibacterial drug based on nanoscale derivatives of chitosan particles

The research presented in the article is related to the hypothesis that the use of an antibacterial drug based on chitosan nanoparticles will increase the safety and effectiveness of the active substance. On the basis of the Department of Therapy and Pharmacology, an antibacterial drug Hit-Line has been developed, where nanoscale chitosan derivatives are used as a carrier for the active substance. The purpose of this study was to study the parameters of subchronic toxicity of an antibacterial drug based on nanomolecular chitosan particles. Based on the goal, the following tasks are set: to evaluate the subchronic toxicity and accumulation of an antibacterial drug based on nanomolecular chitosan particles in laboratory animals with oral administration. All studies were carried out in accordance with the guidelines for conducting preclinical studies of medicines. The cumulative properties of the Hit-Line were studied according to the method of Lim et al. In the first 4 days, 0,1 of LD50 was injected into the stomach, increasing the dose every 4 days by one and a half times. The drug was injected into the body of the test subjects until no more than 1,5 out of 5 animals remained alive in the laboratory group. When determining the parameters of subchronic toxicity, 10 experimental and 3 control groups of white rats with 1 heads each were formed. The drug was administered orally by means of a probe at a dose of 10, 430, 215 mg / kg daily for 86 days. When studying the accumulation of the drug in rats with oral administration, the value of Kk = 28 was established, which corresponds to drugs with pronounced accumulation. As a result of the study of subchronic toxicity in rats, it was found that repeated use of the drug Hit-Line at doses of 1,22, 430 and 215 mg/kg does not cause changes in the clinical status, hematological and biochemical picture of the blood of experimental rats.
