Purification of genetic material concentration from dental samples using EBiS assisted with MNPS
Abstract The purification and characterization of genetic materials are essential in forensic and biomedical applications. Electrical Bioimpedance Spectroscopy (EBiS) assisted with Magnetic Nanoparticles (MNPs) presents a promising approach for non-invasive DNA analysis. This study investigates the use of MNPs in multi-frequency bioimpedance measurements to correlate EBiS with DNA concentrations and its purity. DNA was extracted from dental samples and purified using MNPs, enhancing efficiency of isolation. EBiS measurements were performed before and after purification with a ScioSpec ISX-3 system, using a bipolar electrodes configuration within a 100 Hz to 10 MHz frequency range. DNA concentration and purity were assessed using UV-Vis spectroscopy, and statistical analysis determined the correlation between bioimpedance parameters and genetic material concentration. The results indicate a positive correlation between reactance and DNA concentration at specific frequencies, with an observable alignment trend despite a moderate correlation coefficient. The purification process enhanced DNA quality while maintaining consistent bioimpedance responses, reinforcing the potential of EBIS-assisted methodologies. This study demonstrates the feasibility of EBiS with magnetic nanoparticles for DNA purification and characterization.
