An overview on mucoadhesive drug delivery system

Mucoadhesive drug delivery systems are designed to interact with the mucus layer covering mucosal epithelial surfaces, enhancing the residence time of the dosage form at the absorption site. This interaction facilitates controlled drug release, leading to improved therapeutic outcomes. Over recent decades, mucosal drug delivery has gained significant attention due to its potential benefits, especially for drugs that are susceptible to degradation in the gastrointestinal tract or undergo extensive first-pass metabolism. In recent research, novel poly (ethylene glycol) (PEG)-based mucoadhesive polyesters have been developed by incorporating unsaturated or sulfhydryl functionalities into the polymer backbone, with fluorescein used as a model drug. These modifications aim to enhance the mucoadhesive properties and controlled drug release characteristics of the polymers. Mucoadhesive buccal films have emerged as a promising drug delivery system, offering advantages such as improved patient compliance and the ability to bypass first-pass metabolism. These films adhere to the buccal mucosa, facilitating both local and systemic drug delivery.
