Practical and unified synthesis of prenylated phenylpropanoids via O-reverse prenylation/Claisen rearrangement

Abstract A practical and unified synthetic strategy has been developed for the efficient preparation of bioactive prenylated phenylpropanoids, including drupanin, baccharin, artepillin C, and culifolin. The method employs sequential O-reverse prenylation/Claisen rearrangement, enabling precise control over the number of prenyl groups introduced. The overall process minimizes chromatographic purification and enables gram-scale synthesis in high yield and purity (>95%). This approach provides a scalable platform for synthesizing structurally diverse prenylated phenylpropanoids and facilitates further biological exploration of these underutilized natural products.
