Holistic control strategy: quality and validation as business drivers

Introduction: Process validation is "the collection and evaluation of data, from the process design stage through commercial production, which establishes scientific evidence that a process can consistently deliver a quality product".In a Pharma 4.0 model, a Holistic Control Strategy (HCS) integrates Quality System and the Supply Chain Network (SCN) covering all GxP business processes and several control strategies to ensure quality, safety, efficacy, and data integrity. Here, data is a key asset, enabling a datadriven approach that optimizes manufacturing, ensuring continuous lifecycle feedback. Objectives: To establish the HCS as a comprehensive framework that integrates quality, validation, and the entire SCN into a strategic business model intrinsically linked to the product lifecycle driving innovation, operational agility, and competitive advantage for new product entries.Methodology: A review of 42 ISPE guidelines, 79 PDA guidelines, international regulations (including ICH), and scientific articles was conducted.Risk-based approaches and lifecycle management principles were examined to determine which strategies are necessary and how to integrate them into a holistic control framework for new products as business drivers.Emphasis was placed on digital transformation, adaptive compliance, and the role of advanced analytics in optimizing validation and quality assurance for multidimensional data. Results: Several control strategy models were identified. At least eight strategies were identified and assessed for their impact on new product development across the entire lifecycle within the SCN.Key elements were the application of a risk assessment and digital tools to determine which activities should follow a quality-by-phase approach, which steps need to be streamlined, and how these strategies are interwoven.Process and data flow mapping were identified as vital to aid HCS and to define key activities for each clinical phase, ensuring a structured and data-driven approach to making decisions and breaking the organizational silos to avoid duplicated efforts and suboptimal outcomes. Conclusion:This work marks an initial step in establishing a strategic framework that fosters innovation, accelerates process and product development, and enables data-driven approaches such as Validation 4.0.Building HCS will position quality and validation activities as business drivers, providing a comprehensive view to supporting project, product, and senior managers.Technological investment in digital tools plays a crucial role in integration to reach these goals, ensuring real-time data availability for Data-driven decision-making.
