Anionic surfactant-enhanced ionic liquid-assisted aqueous two-phase microextraction of phenolic acids and flavonoids in the solid matrix from medicinal crop

A novel surfactant-enhanced ionic liquid-assisted aqueous two-phase microextraction (SE-IL-ATPME) method was employed for the efficient extraction and enrichment of phenolic acids and flavonoids in the solid matrix from medicinal crop. The aqueous two-phase system consists of [C 6 MIM]Br, sodium dodecyl sulfate, and NaH 2 PO 4 . The method was systematically optimized by investigating the concentration of ionic liquid, vortex time, amount of salt, amount of SDS, and pH value. The multi-factor experiment was utilized to determine optimal extraction conditions, which were as follows: 146.529 mM of [C 6 MIM]Br concentration, 0.626 g of NaH 2 PO 4 , and 134.013 s of vortex time. The methodology validation demonstrated that the SE-IL-ATPME method achieved excellent linearity (R² > 0.9998), the limits of detection were 0.0164–0.0364 µg/mL, and satisfactory precision of 0.006–0.605 % for retention time and 0.693–3.243 % for peak area. In addition, the SE-IL-ATPME method demonstrated acceptable reproducibility (RSD% < 3.75) and satisfactory accuracy (recoveries ranged from 86.64 % to 108.84 %) in real perillae folium sample analysis. In conclusion, the proposed SE-IL-ATPME method is an efficient, environmentally friendly and rapid method, and has been successfully used to extract phenolic acid phenolic acids and flavonoids from perillae folium. • Anionic surfactant was used to enhance aqueous two-phase system. • A miniaturized aqueous two-phase system was proposed for medicinal crop. • The method was applied to extract phenolic acids and flavonoids. • This approach was validated to be efficient and environmentally friendly.
