8-Hydroxy-2’-deoxyguanosine level and antioxidant status in the dynamics of physiological pregnancy

Background. Due t o the adaptiv e proc esses occurring in a woman’s body during pregnancy, the antioxidant status may be the subject for active changes. An imbalance between antioxidants and prooxidants may lead to the development of oxidative stress. The aim. T o assess of antioxidant status and 8-hydroxy -2’- deoxyguanosine content in the dynamic s of physiological pregnancy . Ma t erials and methods. A prospective study of 32 healthy pregnant women (mean age 28.32 ± 5.21 years) was conducted. Blood plasma and serum, as well as hemolysate prepared from erythrocytes, were used as study material. A comprehensive assessment of the antioxidant status was carried out based on the determination of the level of total antioxidant activity of the blood, the content of reduced and oxidized glutathione and their ratios, glutathione-S-transferase, alpha-tocopherol, retinol and superoxide dismutase activity. Oxidative modification of DNA was assessed by the level of 8-hydroxy-2’-deoxyguanosine. The indicators were studied at 4 points: 6-12, 18-22, 28-32, 38-40 weeks of pregnancy. R esults. It was found that the concentration of retinol, alpha-tocopherol, activity of superoxide dismutase and glutathione-S-transferase significantly increased at 38–40 weeks compared to the first trimester of pregnancy, against the background of a decrease in the level of 8-hydroxy-2’-deoxyguanosine before delivery. Conclusions. The obtained r esults of the study indicat e the activation of the antioxidant defense system in the dynamic s of physiological pregnancy , which leads t o a decrease in oxidative modification of DNA.
