Experiment design: Design and Immunogenicity Study of a Nanoparticle Vaccine Targeting Phosphorylated Tau in Alzheimer's Disease
The Alzheimer's disease Tau hyperphosphorylation hypothesis proposes that in Alzheimer's disease microtubule structure is disrupted and abnormal Tau phosphorylation results in neurofibrillary tangle formation, which is a critical element in the etiology of Alzheimer's disease. The focus of this study is to design a nanoparticle vaccine for Alzheimer's disease (AD) comprising ferritin as a carrier and a polyphosphorylation site pTau as an epitope. By using the SpyTag/SpyCatcher system, a specific and efficient ligation system, we construct and purify a novel nanoparticle protein vaccine. The immunogenicity and safety of the vaccine should then be evaluated in wild type mice.
