MiR-223-3p Positively Regulates Sepsis-Induced Inflammatory Responses by Targeting the Ubiquitin Ligase FBXW7

A growing amount of research is showing that miRNA plays a critical role in the pathophysiology of sepsis. Nevertheless, it is still unclear how miR-223-3p regulates sepsis. The purpose of this study is to look into the relationship and underlying mechanism between sepsis and miR-223-3p. We used qRT-PCR to assess RNA expression. Cell apoptosis was detected employing flow cytometry. The relationship between miR-223-3p and its targeted protein was investigated utilizing luciferase reporter assays. Protein expression was analyzed through western blot and immunofluorescence, while cell viability was determined using the CCK-8 assay. The results showed that miR-223-3p was significantly up-regulated in septic patient serum and LPS-induced RAW264.7 cells, while the expression of FBXW7 showed the opposite pattern. Knockdown of miR-223-3p resulted in a decrease in Bax and cleaved caspase-3 expression, as well as an increase in Bcl-2 expression in LPS-induced RAW264.7 cells. Conversely, knockdown of FBXW7 reversed the effects observed upon miR-223-3p inhibition. Our data demonstrate that MiR-223-3p positively regulates sepsis-induced inflammatory responses by blocking FBXW7 expression, and thus it may provide a new diagnostic marker for sepsis patients.
