From Fresh to Frozen and Everything in Between: Revolutionizing WBC Isolation from Leukopak and Whole Blood with the Rotea Counterflow Centrifugation System

The cell therapy market today comprises numerous companies and academic institutions utilizing various starting materials to produce final cell therapy products. As the cell therapy market continues to grow rapidly, the demand for flexible and efficient processing instruments is increasingly critical. The Rotea Counterflow Centrifugation System is a versatile cell processing instrument capable of isolating white blood cells (WBCs) from multiple sources, including whole blood, fresh leukopak, and frozen leukopak, with minimal viability loss and exceptional cell recovery. The counterflow centrifugation system was used to isolate the WBCs using the 3 different source material with multiple donors to account for the donor to donor variation. The cell recovery and viability pre and post processing was analyzed using an automated counting instrument and the phenotype pre and post processing was measured using flow cytometry. Cells post isolation were also cultured to make sure of no adverse effect in the expansion process. For fresh leukopak, the system offers three distinct methods to isolate WBCs, employing different reagents. These methods cater to customers using legacy reagents like Ficoll and introduce new approaches with ACK lysis buffer for RBC lysis and WBC isolation, achieving a cell recovery rate of 91%. Phenotypic analysis before and after processing indicates no significant differences. For users preferring frozen leukopaks due to their convenience, the system provides an efficient option for WBC isolation, achieving an 85% cell recovery rate without significantly impacting viability. The system also supports whole blood processing, appealing to those seeking time, cost, and resource efficiency. This method yields a 70% cell recovery rate with minimal RBC carryover and no significant viability impact. Phenotypic data confirm no notable differences pre- and post-processing. The process can also be customized to enrich a specific set of cell type as well. In conclusion, the adaptable and modular design of the Rotea Counterflow Centrifugation System makes it an invaluable tool for laboratories aiming to optimize WBC isolation processes. As demand for advanced cell therapy solutions increases, the need for such versatile and efficient instruments will become even more pronounced, ensuring high-quality outcomes in the evolving cell therapy landscape.
