ASSOCIATION OF C.677C>T OF MTHFR GENE WITH INCIDENCE OF MISSED ABORTION IN BABYLONIAN PATIENTS WOMEN

Missed abortion is a common multifactorial disease that usually occurs when an embryo or fetus dies, but the body does not recognize the pregnancy loss.To determine whether the presence of a mutation in the methyl tetrahydrofolate reductase (MTHFR) gene correlates with missed abortions.Randomly of 60 samples were collected for this study and divided into two group, the first one include 30 patient women aged from 20 to 35 years with history of one or more missed first trimester abortions, the second group (control group) which consist of 30 healthy pregnant women who delivered healthy babies (no history of missed abortion ).The DNA was extract from fresh blood and frozen at -20C until PCR begin.PCR product was digested by Hinf1 restriction enzyme and electrophoresis at 2% agarose gel.The results of PCR-RFLP in this study showed increasing significantly the percentage of mutant CT genotype of patients group compared with apparently healthy control, and the allele frequency revealed that T allele was increased significantly in patient group compared with apparently healthy group. This study concluded that both mutant genotype and mutant allele of c.677 C >T in MTHFR gene were associated with incidence of missed abortion in Babylonian patients women.
