PREPARE A NEW STAIN INSTEAD OF EOSIN FROM EXTRACT SEEDS OF RHUS CORIARIA FOR STAINING ANIMAL TISSUES

Sumac plant seeds were employed in preparing aqueous and alcoholic extract for staining animal tissue instead of eosin in a modified way. Methods: the stain was prepared in three methods involving: aqueous sumac solution, aqueous solution containing potassium hydroxide and an alcoholic solution in 70% concentration containing acetic acid. The solutions were used in the staining of different histological organs sections (chicken trachea, liver, kidneys of cows, sheep intestines), and the staining method was modified by the presenting the eosin staining first then haematoxylin the staining step with an increase the time of hematoxylin for approximately 20 minutes. Results: The study showed that the tissue sections responded to the new stain solutions in their three methods, and when they were stained, they gave some results that are better compared to the hematoxylin-eosin stain. In the aqueous solution of sumac, which was used in staining liver tissue, pyknosis was distinguished in the nuclei and the bile duct branch in the portal region. As for the aqueous solution that potassium hydroxide was added to it, the infiltrated inflammatory cells was distinguished better than in eosin stain. The hyaline cartilage could be distinguished clearly in the trachea of sheep, also gastric and goblet cells in the intestine. When using 70% alcoholic sumac solution at a concentration of 20% sumac seeds in staining kidneys sections, the urinary tubes could be distinguished according to their basicity or acidity of cytoplasm.
