Optimisation Of Insitu Gel Formulation Using Central Composite Design by Design Expert Software and Invivo Studies of Delavirdine Nasal Insitu Gel Formulation

The optimisation of Delavirdine formulation was done using central composite design using the design expert software. The critical method parameters like % Drug w.r.t Lipid (Drug: Lipid), Concentration of emulsifier, Pressure in bar with critical Quality parameters Particle Size, PDI, % Drug Entrapment the results show the optimisation of Delavirdine formulation was done using central composite design using the design expert software. The critical method parameters like % Drug w.r.t Lipid (Drug: Lipid), Concentration of emulsifier, Pressure in bar with critical Quality parameters Particle Size, PDI, % Drug Entrapment the results show Quadratical Model with Parameters Predicted value and Observed Values Particle size 155.31and 155.7, PDI 0.367356              and 0.37 Entrapment Efficacy 94.1452 and 94.57. It was observed that the differences between the observed and predicted peak response were less than 5%. In vivo studies were performed for optimised intra nasal formulation the bioavailable of Plasma and brain of the optimised formation was calculated found to be 84.4 % with the developed formulation while it was found to be 92.3 % with the Marketed Drug. Relative bioavailability was determined to be with the developed formulation indicating slight Lesser absorption potential of the Delavirdine loaded SLN dispersion in comparison to the orally administered drug powder.
