MOLECULAR CHARACTERIZATION OF BACTERIA ISOLATE FROM SOIL OF CATTLE KRAAL IN OYO STATE COLLEGE OF AGRICULTURE AND TECHNOLOGY, IGBOORA

Soil serves as a host for diversity of many organisms with great potential to reform and to destroy. The soil bacteria were isolated by standard serial dilution plate techniques. Morphological characterization of the isolate was done by Gram’s staining and biochemical characterization. Molecular characterization was conducted by Deoxyribonucleic acid (DNA) extraction and sequencing one of the bacteria isolates. Three Gram positive and three Gram-negative bacteria were isolated from this experimentation involving six genera. The frequency of occurrence of the organisms are as follows: Bacillus cereus is 25%, Bacillus subtilis and Clostridium spp are 16.7%, Bacillus spp, Staphylococcus aureus, Enterobacter spp, Neisseria spp and Pseudomonas spp are 8.3%. The DNA sequencing of isolate suspected to be Pseudomonas has 90.81% pair wise similarity with Pseudomonas pseudoalcaligenes strain SG-2 which has National Center for Biotechnology Information (NCBI) accession number JQ04008. Isolates from this study has potential to damage i.e. cause harm to our livestock and protect them. More studies should be conducted on maximizing the potential of the protective organism.
