Advances in formulation strategies and stability considerations of amorphous solid dispersions
The increasing prevalence of poorly water-soluble drugs highlights the need for effective formulation strategies, with amorphous solid dispersions (ASDs) offering a promising solution to enhance solubility and bioavailability. This review consolidates existing knowledge, examine key advances, and delivers a broad yet well-structured perspective on the field. It explores the physicochemical properties of drugs and their impact on formulation strategies, examines the role of polymers in maintaining supersaturation and inhibiting crystallization , and addresses stability challenges, including molecular mobility, drug-polymer interactions, and environmental influences such as moisture. Advanced characterization techniques are reviewed for their predictive value in ensuring long-term stability. Additionally, the regulatory and commercial landscape underscores ASDs’ potential to address unmet medical needs. By bridging scientific innovation with practical application, ASDs continue to shape pharmaceutical science and drive progress toward next-generation drug delivery systems .
