Bioactive homoisoflavonoids from the fibrous roots of Ophiopogon japonicus

Phytochemical exploration of the fibrous roots of Ophiopogon japonicus culminated in the identification of two novel homoisoflavonoids , designated as 5- O -methylophiopogonone C ( 1 ) and (3 S )-7,2’-dihydroxy - 3’,4’-methylenedioxyhomoisoflavan ( 2 ). Leveraging an array of advanced spectroscopic methodologies, inclusive of nuclear magnetic resonance (NMR), infrared spectroscopy (IR), ultraviolet spectroscopy (UV), and high - resolution electrospray ionization mass spectrometry (HRESIMS), the structures of two isolated compounds were meticulously elucidated. Subsequently, their tyrosine kinase inhibitory activities were evaluated. • Two new homoisoflavonoids, namely as 5- O -methylophiopogonone C ( 1 ), and (3 S )-7,2’-dihydroxy-3’,4’-methylenedioxyhomoisoflavan ( 2 ) were isolated and structural elucidated. • Compounds featuring the structural characteristics of 2, a C2-C3 single bond and lacking the C-4 carbonyl group (replaced by two hydrogens), have been isolated from the genus Ophiopogon for the second time. • Compound 2 exhibited a moderate tyrosinase inhibitory activity (IC 50 27.6 μg/mL), suggesting that it might potentially serve as a candidate for the treatment of skin diseases related to melanin biosynthesis.
