Isolation and Extraction of Gentian Polysaccharide and Its Protective Effect on Chemical Liver Injury

ABSTRACT This study aimed to isolate Gentian polysaccharide and explore the protective effects of Gentian polysaccharide on lipopolysaccharide combined with D‐galactosamine (D‐GaLN/LPS) induced chemical liver injury, theoretical and material basis for the prevention and treatment of liver injury. A water‐soluble polysaccharide (GSBP‐N‐a‐1), with an average molecular weight of 3.82 kDa, was isolated and purified from the water extract of Gentian using Diethylaminoethyl‐cellulose (DEAE) column chromatography, CL‐6B agarose gel chromatography, and Sephadex G25 chromatography. A chemical liver injury model was established in mice via intraperitoneal injection of D‐GalN/LPS. In vivo experiments showed that Gentian crude polysaccharide had a protective effect on liver injury models induced by lipopolysaccharide combined with D‐galactosamine. It may act by activating the Nrf2‐HO‐1 pathway to inhibit oxidation and reduce the release of inflammation.
