Regorafenib in Caucasian patients with advanced KIT-mutant melanoma: a dual center case series – Updated results
discontinuation, 19 of 35 patients (54%) experienced disease progression, with 13 of 19 patients (68%) progressing within the first 12 months.Reinduction with BRAFi/MEKi in 17 of 19 progressing patients resulted in a 76% objective response rate, while mPFS post-reinduction was not reached. Conclusions: In this real-world national cohort of melanoma patients with long-term responses to BRAFi/MEKi, a sizable proportion of patients discontinuing treatment without progressive disease became long-term survivors.Progression after discontinuation typically occurred within the first year, with reinduction of BRAFi/MEKi being highly effective.Further research is needed to identify clinical or molecular features predictive of sustained tumor control post-discontinuation to refine treatment guidelines.
