Early-stage autophagy inhibitors as a therapeutic target in BRAFV600E melanoma cells
Background: Melanoma is a malignant neoplasm that is highly resistant to chemotherapy and radiotherapy and is associated with a poor prognosis in advanced stages. BRAFV600E inhibitors (BRAFi), such as Vemurafenib, reduce tumor burden, but these tumors frequently acquire resistance to these drugs. In cancer, autophagic flux acts as a survival mechanism that allows the tumor to handle with increased metabolic demand and damage caused by chemotherapeutics. Melanoma cells show constitutive activation of autophagy.
